Core Facility Viral Vector and Genome Editing Technologies

The Vector-Core-Facility is a non-profit service institution at the IEGT. The core facility supports basic research at Rostock University Medical Center and Rostock University. Upon request, external partners can benefit from our services. The facility produces for research purposes custom-made viral or non-viral gene transfer systems, so-called vectors. Vectors allow the introduction of genetic material into eucaryotic cells or laboratory animals. The genetic material may be designed to overexpress proteins or RNAs or to selectively silence the activity of a single cellular gene. In addition, the use of CRISPR/Cas9 technology offers the possibility to make targeted genetic modifications in different genomes (genome editing). The suitable vector system will be selected depending on the scientific question. For transient genetic alterations, adenoviral-derived vectors are the tools of choice. Vectors on the basis of lentiviruses allow long-term expression of a transgene. State-of-the-art vector technologies can compensate for transgenic mice or knockout mouse models. In this respect, our technology platform has a strong focus on the development cell- or tissue-specific vectors (so-called SmartViruses) for systemic applications.

The service of the Vector-Core-Facility also includes

  • upfront consultation within a research project concerning the vector system(s) of choice,
  • support in planning vector-related scientific experiments;
  • cloning of the gene of interest or shRNA into the viral vector of choice, its production, amplification, titration and initial testing;
  • cloning of the gene of interest or shRNA into the viral vector of choice, its production, amplification, titration and initial testing;
  • scientific and technical training for local users, students and guest scientists;
  • counselling and assistence with safety requirements and preparation of paperwork to comply with national legal regulations (Germany).
